Reporter Genes02:11

Reporter Genes

Reporter genes are a type of protein-coding gene that are often tagged to a gene of interest. Once inside a target cell, reporter genes usually produce visually identifiable characteristics like fluorescence and luminescence when expressed along with the gene of interest. Thus, reporter genes “report” the presence or absence of genes of interest in an organism, determine the gene expression pattern, or track the physical location of a DNA segment or protein in the cell.

Summary

We developed the mT/mG mouse, a novel double-fluorescent Cre reporter. This tool enables precise tracking of Cre-mediated recombination and cell morphology across all tissues in live and fixed samples.

Area of Science:

  • Genetics and Genomics
  • Molecular Biology
  • Developmental Biology

Background:

  • The Cre/loxP system is crucial for conditional mutagenesis in mice.
  • Accurate reporters of Cre activity are essential for understanding spatial and temporal recombination events.

Purpose of the Study:

  • To introduce mT/mG, a double-fluorescent Cre reporter mouse model.
  • To evaluate the reporter's utility for visualizing Cre activity and cell morphology.

Main Methods:

  • Development of a mouse line expressing membrane-targeted tandem dimer Tomato (mT) and membrane-targeted green fluorescent protein (mG).
  • Assessment of reporter expression across various tissues in live and fixed samples.
  • Validation of Cre-dependence, single-cell resolution, and FACS distinguishability of mG labeling.

Main Results:

  • mT/mG reporter expression is nearly ubiquitous across all examined tissues.
  • mG labeling is strictly Cre-dependent and complementary to mT at the single-cell level.
  • Membrane-targeted fluorescent proteins effectively outline cell morphology and visualize cellular processes.

Conclusions:

  • The mT/mG mouse serves as a versatile global Cre reporter.
  • This reporter system facilitates lineage tracing, transplantation studies, and in vivo cell morphology analysis.
