Area of Science:

  • Biomedical Engineering
  • Cardiovascular Research
  • Respiratory Support Devices

Background:

  • The MC3 Biolung total artificial lung (TAL) underwent 30-day testing in preparation for clinical trials.
  • This study evaluated the performance and safety of the TAL in a preclinical ovine model.

Purpose of the Study:

  • To assess the feasibility and performance of the MC3 Biolung total artificial lung (TAL) for extended respiratory support.
  • To identify potential areas for improvement in device biocompatibility and anticoagulation strategies.

Main Methods:

  • The TAL was implanted in 8 sheep, with hemodynamic and blood gas parameters monitored regularly.
  • Device performance was assessed by monitoring resistance and oxygen transfer, with replacement criteria defined.
  • Organ function and post-mortem findings were evaluated after 30 days or premature euthanasia.

Main Results:

  • Five of eight sheep survived the 30-day study period, with survivors exhibiting stable hemodynamics and blood gases.
  • The TAL demonstrated consistent oxygen transfer and acceptable resistance levels throughout the study.
  • Increased platelet and white blood cell counts were observed, with mild ischemic kidney lesions noted at necropsy.

Conclusions:

  • Thirty-day respiratory support using the MC3 Biolung TAL is feasible in an ovine model.
  • Improvements in biocompatibility and anticoagulation are necessary to mitigate device-related thrombogenicity.
