Glycosaminoglycans (GAGs), also known as mucopolysaccharides, are long and linear polymers comprising of specific repeating disaccharides - the amino sugar that can be N-acetylglucosamine or N-acetylgalactosamine, and a uronic acid that is usually glucuronic acid or iduronic acid.

  • Primary rabbit articular chondrocytes were cultured and treated with IL1β, CS, or both.
  • Newly synthesized PGs were labeled with [35SO4]-sulphate and analyzed using Sepharose 2B chromatography.
  • Gene and protein expression of matrix metalloproteinases (MMPs), ADAMTS-4, and ADAMTS-5 were quantified via real-time PCR and Western blotting.

Main Results:

  • CS significantly increased the production of sulphated PGs within the cellular pool.
  • CS partially inhibited the IL1β-induced disaggregation of sulphated PGs.
  • This protective effect correlated with a significant reduction in ADAMTS-5 expression at both mRNA and protein levels.
