Summary

Recombinant human serum albumin (rHSA) offers a safe and effective alternative to plasma-derived HSA for maintaining oncotic pressure. Produced in yeast, rHSA is highly pure and functionally equivalent to pdHSA.

Area of Science:

  • Biotechnology
  • Biochemistry
  • Medical Science

Background:

  • Human serum albumin (HSA) maintains plasma colloid osmotic pressure, crucial for fluid balance.
  • Clinical applications often require large doses (>10g), increasing risks associated with plasma-derived HSA (pdHSA) contamination.
  • Recombinant HSA (rHSA) production offers a potential solution to pdHSA safety concerns.

Purpose of the Study:

  • To evaluate the safety and efficacy of a recombinant human serum albumin (rHSA) preparation.
  • To confirm the structural and functional similarity of rHSA to plasma-derived HSA (pdHSA).
  • To assess rHSA's suitability as a plasma expander and biomaterial.

Main Methods:

  • rHSA was produced using the methylotrophic yeast Pichia pastoris.

  • Purification was achieved using Streamline technology.
  • Structural and functional properties were compared to pdHSA.
  • Preclinical and clinical trials were conducted.
  • Main Results:

    • rHSA purified by Streamline technology showed no detectable mannan component.
    • Structural and functional analyses confirmed rHSA is identical to pdHSA.
    • Preclinical and clinical trials demonstrated the safety and efficacy of rHSA.
    • rHSA proved effective in conditions like hemorrhagic shock and cirrhosis with ascites.

    Conclusions:

    • The Pichia pastoris-derived rHSA is a safe and effective substitute for pdHSA.
    • rHSA maintains essential oncotic pressure and plasma volume.
    • rHSA demonstrates significant potential beyond plasma expansion, including use as a biomaterial.
