DNA Microarrays02:34

DNA Microarrays

Microarrays are high-throughput and relatively inexpensive assays that can be automated to analyze large quantities of data at a time. They are used in genome-wide studies to compare gene or protein expression under two varied conditions, such as healthy and diseased states. Microarrays consist of glass or silica slides on which probe molecules are covalently attached through surface functionalization. Most commonly, the slides are prepared through the chemisorption of silanes to silica...

Area of Science:

  • Genomics
  • Molecular Biology
  • Bioinformatics

Background:

  • High-throughput sequencing requires efficient methods for targeting specific genomic regions.
  • Previous methods like multiplex PCR have limitations for large-scale genomic analysis.

Purpose of the Study:

  • To evaluate the efficacy of high-density microarrays for enriching specific human genomic sequences.
  • To compare microarray-based enrichment with multiplex PCR for large-scale genomic region analysis.

Main Methods:

  • Application of high-density microarrays for capturing 6,726 exon segments (approx. 500 bases each).
  • Capture of locus-specific genomic regions (200 kb to 5 Mb).
  • Sequencing of captured targets using a 454 Life Sciences FLX sequencer.

Main Results:

  • The majority of sequence reads obtained directly corresponded to the targeted genomic regions.
  • Microarray-based enrichment demonstrated high specificity for both exon and locus-specific targets.
  • Successful sequencing of enriched targets was achieved.

Conclusions:

  • High-density microarrays provide an effective method for targeted enrichment of human genomic DNA.
  • This direct selection approach is superior to multiplex PCR for large-scale genomic analyses.
  • The method facilitates efficient high-throughput sequencing of selected genomic regions.
