Real Time RT-PCR02:57

Real Time RT-PCR

Real-time reverse transcription-polymerase chain reaction, or Real-time RT-PCR, is an analytical tool used to determine the expression level of target genes. The method involves converting mRNA to complementary DNA with the help of an enzyme known as reverse transcriptase, followed by the PCR amplification of the cDNA. These two processes can be performed simultaneously in a single tube or separately as a two-step reaction.
The real-time quantification of the number of amplified products is...

Summary
This summary is machine-generated.

Prostate cancer antigen 3 (PCA3) shows low positivity in circulating cells of prostate cancer patients. This study developed a new assay, finding no PCA3 expression in healthy individuals, unlike prostate-specific antigen (PSA).

Area of Science:

  • Molecular diagnostics
  • Oncology
  • Biomarker discovery

Background:

  • Prostate cancer antigen 3 (PCA3) is a non-translated gene specific to the prostate.
  • PCA3 is known to be overexpressed in prostate cancer tissues.

Purpose of the Study:

  • To develop a quantitative, internally standardized real-time RT-PCR assay for PCA3.
  • To investigate the presence and role of PCA3 in the peripheral blood of prostate cancer patients.

Main Methods:

  • A real-time RT-PCR assay utilizing target-specific lanthanide probes was developed for PCA3 detection.
  • Peripheral blood samples from 91 patients with prostate cancer/disorders and healthy controls were analyzed for PCA3 and prostate-specific antigen (PSA) expression.

Main Results:

  • The developed PCA3 assay demonstrated a wide dynamic range (over eight orders of magnitude) and a low limit of quantification (800 copies/mL).
  • PCA3 was detected in circulating cells of 2 out of 9 patients with metastasized prostate cancer; all other samples were negative.
  • Prostate-specific antigen (PSA) was detected in eight samples.

Conclusions:

  • PCA3 positivity in circulating cells of prostate cancer patients is less frequent compared to PSA.
  • Unlike some prior studies, this research found no detectable PCA3 expression in healthy individuals, suggesting its potential as a specific marker.
