A convergent total synthesis of (+)-yatakemycin was achieved in 20 steps. New methods for aryl-nitrogen bond formation and debenzylation enabled subgram-scale production of this complex molecule.
Area of Science:
- Organic Synthesis
- Medicinal Chemistry
- Total Synthesis
Background:
- Yatakemycin is a complex natural product with potential biological activity.
- Efficient synthetic routes are crucial for accessing and studying such molecules.
- Previous syntheses may have faced challenges in stereocontrol or bond formation.
Purpose of the Study:
- To develop a convergent total synthesis of (+)-yatakemycin.
- To establish novel methodologies for key bond formations and functional group manipulations.
- To achieve a practical, subgram-scale synthesis of the target molecule.
Main Methods:
- Convergent synthesis strategy employing a 20-step sequence.
- Regioselective ring opening of (S)-epichlorohydrin to install chirality.
- Copper-catalyzed aryl amination (CuI/CsOAc) for constructing five aryl-nitrogen bonds.
- Chemoselective debenzylation using BCl3 and pentamethylbenzene.
Main Results:
- Successful completion of the total synthesis of (+)-yatakemycin in 13% overall yield.
- Development of efficient and stereoselective methods for key synthetic steps.
- Demonstration of subgram-scale synthesis feasibility.
Conclusions:
- The developed synthetic route provides a viable pathway to (+)-yatakemycin.
- The novel methodologies employed are broadly applicable to complex molecule synthesis.
- This work facilitates further investigation into the properties and applications of yatakemycin.
