Background:

Avermectin and its analogs are crucial antiparasitic agents in veterinary, agricultural, and human medicine.
Understanding the genetic basis of avermectin overproduction is vital for improving commercial yields.

Purpose of the Study:

To elucidate the genetic mechanisms behind avermectin overproduction in Streptomyces avermitilis.
To compare gene expression profiles between low and high avermectin-producing strains.

Main Methods:

Comparative transcriptome analysis using a whole genome chip.
Identification of differentially expressed genes between S. avermitilis ATCC31267 (low producer) and S. avermitilis ATCC31780 (high producer).

Main Results:

Fifty S. avermitilis genes showed at least two-fold higher expression in the high-producing strain.
Avermectin biosynthetic genes, including polyketide synthase (PKS) and regulatory genes, were underexpressed in the low-producing strain.
The identified fifty genes represent potential novel contributors to avermectin overproduction.

Conclusions:

Avermectin overproduction in S. avermitilis ATCC31780 is associated with the upregulation of fifty previously unidentified genes.
Increased transcription of avermectin PKS genes also contributes to higher avermectin yields.
These findings provide insights into the genetic regulation of avermectin biosynthesis.
