Summary

Split ejaculation separates motile sperm for fertility treatments. In vasovasostomized men, antisperm antibodies are present throughout the ejaculate, suggesting serum transudation from multiple reproductive tract levels, including seminal vesicles.

Background:

  • Split ejaculation technique is utilized to concentrate motile spermatozoa for assisted reproductive technologies.

Purpose of the Study:

  • To investigate the distribution and origin of antisperm antibodies in the split ejaculate of vasovasostomized men.

Main Methods:

  • Mixed antiglobulin reaction (MAR) test for IgG, IgA, and secretory component on whole ejaculate and split fractions.
  • Indirect MAR and tray agglutination test to determine free ASA isotype and concentration in serum and ejaculate.

Main Results:

  • Bound antisperm antibodies showed similar reactivity in the first two fractions and the whole ejaculate.
  • Free antisperm antibodies were detected in serum and all ejaculate fractions.

Conclusions:

  • Antisperm antibodies in vasovasostomized men's ejaculate originate from serum transudation.
  • This transudation occurs not only at the epididymal/prostatic levels but also within the seminal vesicles.
