  • Robo family members (Robo1, Robo2, Robo3) have complementary roles in the tract of the postoptic commissure (TPOC).
  • Robo2 mediates initial fascicle splitting in response to Slit1a.
  • Robo1 and Robo3 restrict defasciculation, balancing tract formation.
  • Robo-Slit signaling is essential for vertebrate longitudinal axon guidance.

Conclusions:

  • The Robo-Slit signaling pathway plays a key role in vertebrate longitudinal axon guidance.
  • Complementary functions of Robo receptors are vital for precise axon navigation within complex pathways.
