    Summary

    Chronic myelogenous leukemia (CML) leukocytes show enhanced interferon production. This process requires new protein synthesis and is not affected by cytostatics, but is inhibited by RNA or protein synthesis blockers.

    Background:

    • Interferon (IF) production is a key immune response to viral infections.
    • Leukocyte function can be altered in patients with hematological malignancies like chronic myelogenous leukemia (CML).

    Purpose of the Study:

    • To investigate the in vitro production of interferon (IF) induced by Sendai virus in human leukocytes from normal donors and CML patients.
    • To determine the impact of cytostatics and inhibitors of nucleic acid/protein synthesis on IF production in CML leukocytes.

    Main Methods:

    • Partially purified human leukocyte suspensions from normal donors and CML patients were used.
    • Leukocytes were stimulated with Sendai virus in vitro.
    • Interferon production was measured in the presence and absence of serum, cytostatics, and inhibitors of RNA/protein synthesis.

    Main Results:

    • Leukocytes from CML patients in exacerbation demonstrated increased IF production compared to normal leukocytes, while those in remission showed normal production.
    • IF production was unaffected by cytostatics but was inhibited by actinomycin D, puromycin, and cycloheximide, indicating a requirement for de novo protein synthesis.
    • CML leukocytes exhibited prolonged IF production compared to normal leukocytes.
    • Leukocytes from children with acute blastic leukemia and adults with chronic lymphoid or acute paramyeloblastic leukemia produced IF similarly with or without serum, unlike normal leukocytes.

    Conclusions:

    • Sendai virus-induced interferon synthesis in chronic myelogenous leukemia leukocytes is dependent on de novo protein synthesis.
    • Leukocyte IF production capacity is altered in various leukemia subtypes, with CML showing distinct characteristics during exacerbation.
