SNPs in PPARG associate with type 2 diabetes and interact with physical activity

Certain single-nucleotide polymorphisms (SNPs) in the PPARG gene are linked to type 2 diabetes (T2D) risk. Increased physical activity (PA) may mitigate this risk, particularly for specific PPARG gene variants.

- The study analyzed 479 overweight individuals with IGT from the Finnish Diabetes Prevention Study (DPS).
- Participants were followed for an average of 4.2 years.
- Yearly assessments of physical activity (PA) were conducted using a validated questionnaire.
Main Results:
- Two SNPs, rs17036314 and rs1801282 (Pro12Ala), were associated with an increased risk of conversion to T2D.
- The association for rs17036314 remained significant after adjusting for baseline fasting glucose.
- Increased physical activity significantly modified the risk associated with rs17036314 and rs1801282, seemingly negating the risk conferred by the associated alleles.
- A distinct SNP, rs1152003, showed an interaction with the study group, tending to increase T2D risk in the intervention group, but this was not modified by PA changes.
Conclusions:
- Specific PPARG gene SNPs (rs17036314, rs1801282, and rs1152003) are associated with T2D risk in the DPS cohort.
- Higher levels of physical activity appear to counteract the T2D risk associated with rs17036314 and rs1801282.
- The influence of rs1152003 on T2D risk may be modulated by broader lifestyle interventions or other lifestyle factors.
