    Summary

    Corticosteroids like ultracortene and ACTH significantly damage eosinophils in hypereosinophilic syndrome by destroying granules and altering cell structures. Antihistamines showed no such effect.

    Background:

    • Hypereosinophilic syndrome is characterized by excessive eosinophils.
    • Corticosteroids are a common treatment for hypereosinophilic syndrome.

    Observation:

    • Eosinophils from a hypereosinophilic syndrome patient were studied in vitro.
    • The effects of ultracortene, ACTH, and mepyramine were examined.

    Findings:

    • Ultracortene and ACTH caused significant ultrastructural damage to eosinophils.
    • Damage included granule destruction, mitochondrial swelling, and cristae distortion.
    • The antihistamine mepyramine did not induce ultrastructural alterations.

    Implications:

    • Corticosteroids induce significant cellular damage in hypereosinophilic syndrome eosinophils.
    • This provides insight into the mechanism of corticosteroid action.
    • Further research may explore targeted therapies to mitigate these side effects.
