Detailed Structure and Function of Lymph Nodes

Lymph nodes are bean-shaped structures that cluster along the lymphatic vessels in the inguinal, axillary, and cervical regions. Each node is divided into compartments by a capsule that extends trabeculae inward.
From a histological perspective, lymph nodes can be split into two main areas: the superficial cortex and the deep medulla. The outer cortex is populated by dendritic cells, macrophages, and B lymphocytes, which are densely packed into follicles. When these B-lymphocytes are presented...

Summary

Identifying mouse lymph nodes is crucial for cancer and immunity research. New dye injection techniques improve visualization of lymphatic drainage in mice, aiding disease studies.

Area of Science:

  • Immunology
  • Oncology
  • Veterinary Anatomy

Background:

  • Accurate identification of lymph nodes is vital for studying tumor metastasis and immune responses.
  • Standard dissection methods in mice are challenging due to the small size of lymphatic organs.
  • Larger rodents are often used, but mouse-specific lymphatic drainage patterns are important.

Purpose of the Study:

  • To develop and validate techniques for accurate and rapid identification of lymph nodes and lymphatic drainage in mice.
  • To overcome the limitations of standard dissection techniques for small rodent lymphatic organs.

Main Methods:

  • Injection of dye into the mouse footpad or tail to label lymphatic drainage.
  • Characterization of lymphatic drainage for the hind leg, flank, pelvic viscera, prostate, and mammary glands.

  • Comparison of mouse lymphatic drainage patterns with those of rats.
  • Main Results:

    • Developed effective dye injection techniques for visualizing mouse lymphatic systems.
    • Identified specific lymphatic drainage pathways in mice for various anatomical regions.
    • Observed distinct differences in afferent and efferent drainage of inguinal lymph nodes between mice and rats.

    Conclusions:

    • The developed dye injection techniques enable accurate and rapid identification of lymph nodes and lymphatic drainage in mice.
    • These methods are valuable for research on tumor metastasis and regional immune responses in both normal and diseased mice.
    • The findings highlight species-specific differences in rodent lymphatic anatomy, particularly in inguinal lymph nodes.
