Single Nucleotide Polymorphisms-SNPs01:05

Single Nucleotide Polymorphisms-SNPs

A single nucleotide polymorphism or SNP is a single nucleotide variation at a specific genomic position in a large population. It is the most prevalent type of sequence variation found in the human genome. Point mutations that occur in more than 1% of the population qualify as SNPs. These are present once every 1000 nucleotides on an average in the human genome. Replacement of a purine with another purine (A/G) or a pyrimidine with another pyrimidine (C/T) is known as a transition. In contrast,...

Summary

Two novel actinomycete strains, Nocardioides hwasunensis sp. nov., were discovered in Jeju Island, Republic of Korea. These aerobic, Gram-positive bacteria represent a new species within the Nocardioides genus.

Area of Science:

  • Microbiology
  • Bacteriology
  • Taxonomy

Background:

  • Actinomycetes are a diverse group of bacteria with significant ecological and biotechnological roles.
  • The genus Nocardioides is known for its metabolic versatility and presence in various environments.
  • Isolation of novel species is crucial for expanding our understanding of microbial diversity and potential applications.

Purpose of the Study:

  • To isolate and characterize novel actinomycete strains from marine environments.
  • To determine the taxonomic position of the isolates using polyphasic characterization.
  • To propose a new species within the genus Nocardioides.

Main Methods:

  • Isolation of actinomycetes from seawater samples.
  • Phenotypic characterization including morphology, growth conditions, and chemotaxonomic analyses (cell wall, menaquinone, fatty acids, polar lipids).
  • Molecular phylogenetic analyses based on 16S rRNA gene sequencing and DNA-BOX-PCR fingerprinting.

Main Results:

  • Two novel aerobic, Gram-positive, non-motile rod strains, HFW-18 and HFW-21(T), were isolated.
  • Chemotaxonomic data (LL-diaminopimelic acid, MK-8(H(4)), G+C content, fatty acid profile) were consistent with the genus Nocardioides.
  • Phylogenetic analysis based on 16S rRNA gene sequences placed the isolates within the Nocardioides genus, with N. ganghwensis JC2055(T) as the closest relative (97.4% similarity).

Conclusions:

  • The isolates HFW-18 and HFW-21(T) represent a novel species of the genus Nocardioides.
  • The proposed name for the new species is Nocardioides hwasunensis sp. nov.
  • The type strain is HFW-21(T) (=KCTC 19197(T)=DSM 18584(T)).
