Abstract:
The hedgehog (Hh) signaling pathway was originally found as an organizer in embryonic development. The pathway is now implicated in the development of various tumors. Recently, it has been reported that the pathway activation is resulted from aberrant expression of the ligand, Sonic Hh (Shh), in pancreatic cancer and breast cancer. Here we developed a new strategy to control the activation of the Hh signaling pathway in cancer cells, which have ectopic and ligand dependent activation of the Hh signaling pathway. Our strategy may contribute to the development of a new cancer treatment.
Insights
Researchers developed a novel strategy to control the hedgehog (Hh) signaling pathway in cancer cells. This approach targets aberrant pathway activation, potentially leading to new cancer treatments.
Area of Science:
- Oncology
- Developmental Biology
- Molecular Signaling
Background:
- The hedgehog (Hh) signaling pathway is crucial for embryonic development.
- Aberrant Hh pathway activation, often due to Sonic Hh (Shh) ligand overexpression, is implicated in pancreatic and breast cancer development.
Purpose of the Study:
- To develop a novel strategy for controlling Hh signaling pathway activation in cancer cells.
- To target cancers with ectopic and ligand-dependent Hh pathway activation.
Main Methods:
- Development of a new strategy to modulate Hh pathway activity.
- Application of the strategy to cancer cells exhibiting specific pathway activation patterns.
Main Results:
- Successful control of Hh signaling pathway activation in cancer cells was achieved.
- The developed strategy is effective in contexts of ectopic and ligand-dependent pathway activation.
Conclusions:
- The novel strategy offers a potential method for controlling aberrant Hh signaling in cancer.
- This research may pave the way for new therapeutic approaches in oncology, particularly for cancers driven by Hh pathway dysregulation.

However, cancer treatments can pose several challenges, as therapies used to kill cancer cells are generally also toxic to normal cells. Moreover, cancer cells mutate rapidly and can develop resistance to chemical agents or radiation therapy. Besides, all types of cancer cells may not respond to the same therapy. Some cancer cells respond to one...
