    Secretions like tears and saliva in chickens develop strong Newcastle disease virus (NDV) neutralizing activity after ocular infection. This localized immunity, primarily IgA, limits reinfection but doesn't fully prevent it.

    Background:

    • Newcastle disease virus (NDV) is a significant pathogen in poultry.
    • Understanding mucosal immunity is crucial for controlling viral infections in chickens.

    Purpose of the Study:

    • To investigate the development and characteristics of specific antibody responses in ocular and upper respiratory secretions following ocular NDV infection.
    • To evaluate the role of the Harderian gland in mucosal immunity against NDV.

    Main Methods:

    • Specific pathogen-free chickens were exposed ocularly to lentogenic NDV.
    • Virus-neutralizing activity and antibody isotypes (IgA, IgG) in lacrimal fluid, saliva, and tracheal washes were analyzed.
    • Chickens were rechallenged to assess secondary responses in secretions and circulating antibodies.
    • Harderian gland activity was examined histologically, and its function was assessed via duct occlusion.

    Main Results:

    • Ocular NDV infection induced significant virus-neutralizing activity in lacrimal fluid, saliva, and tracheal washes, exceeding circulating antibody levels.
    • IgA was the predominant antibody isotype in secretions, particularly saliva (85%), with IgG also present.
    • Secreted antibodies partially limited, but did not prevent, tracheal reinfection upon challenge.
    • Reinfection induced a primary-like response in secreted antibodies, unlike the elevated circulating antibody titers.
    • Ocular infection stimulated lymphoid and plasma cell activity in the Harderian gland, a key source of antibodies in ocular and other secretions.

    Conclusions:

    • The Harderian gland plays a major role in generating specific antibodies in ocular and upper respiratory secretions following NDV infection.
    • Secreted IgA is a critical component of the initial mucosal immune response to ocular NDV exposure.
    • While contributing to defense, secreted antibodies alone do not confer complete protection against reinfection.
