Background:

  • Ghrelin, a stomach-derived peptide, stimulates appetite.
  • Obestatin, derived from the same precursor as ghrelin, was initially thought to inhibit ghrelin's effects.
  • The postprandial (after-meal) behavior of obestatin and its relationship with ghrelin in humans were not well understood.

Purpose of the Study:

  • To investigate the postprandial response of obestatin, ghrelin (total, acyl, and desacyl forms), and Neuropeptide Y (NPY) after a high-carbohydrate breakfast in healthy women.
  • To understand the interplay between these hormones in regulating appetite after food intake.

Main Methods:

  • Eight healthy women consumed a standardized high-carbohydrate breakfast (1604 kJ).
  • Blood samples were collected at multiple time points (pre-meal, 30, 60, 90, 120, 150 minutes post-meal).
  • Plasma concentrations of obestatin, total ghrelin, acyl ghrelin, desacyl ghrelin, and NPY were measured.

Main Results:

  • Plasma obestatin concentrations significantly decreased post-meal, reaching lowest levels at 90 minutes.
  • Total ghrelin concentrations also significantly decreased, with nadir at 120 minutes.
  • Acyl and desacyl ghrelin levels decreased at 30 and 60 minutes, respectively. NPY levels were lower at 90 and 150 minutes post-meal.

Conclusions:

  • Postprandial plasma obestatin concentrations decrease similarly to ghrelin levels after a high-carbohydrate meal in healthy young women.
  • This suggests a coordinated response of these peptides following nutrient intake.
  • Further research is needed to fully elucidate the roles of obestatin and ghrelin in human appetite regulation.
