Pathologic gamblers show lower levels of cerebrospinal fluid (CSF) 5-hydroxyindoleacetic acid (5-HIAA) and 4-hydroxy-3-methoxyphenyl glycol (HMPG), suggesting altered serotonin and norepinephrine function.

Background:

  • Previous studies indicated disturbed norepinephrine and dopamine function in pathologic gamblers' cerebrospinal fluid (CSF).
  • Serotonin system functioning in pathologic gambling has remained less understood.

Purpose of the Study:

  • To investigate neurotransmitter metabolite levels in the CSF of pathologic gamblers.
  • To compare CSF neurotransmitter metabolite profiles between pathologic gamblers and healthy controls.

Main Methods:

  • Analyzed two CSF samples from 10 male pathologic gamblers and healthy male controls.
  • Measured concentrations of 5-hydroxyindoleacetic acid (5-HIAA), homovanillic acid (HVA), and 4-hydroxy-3-methoxyphenyl glycol (HMPG).
  • Accounted for CSF mass flow rate (tapping time) for concentration calculations.

Main Results:

  • CSF 5-HIAA and HMPG levels, adjusted for tapping time, were significantly lower in gamblers compared to controls.
  • CSF HVA levels did not differ significantly between groups when adjusted for tapping time.
  • A significant correlation between 5-HIAA and HVA was observed in gamblers, but not in controls.

Conclusions:

  • Altered indolamine (serotonin precursor) function in pathologic gamblers cannot be ruled out, despite previous findings focused on norepinephrine and dopamine.
  • Findings suggest potential dysregulation in serotonin pathways contributing to pathologic gambling behavior.
