Background:

  • Cancer-associated stromal cells can acquire copy number alterations and p53 mutations.
  • Pancreatic ductal adenocarcinoma is a complex disease involving tumor cells and their microenvironment.

Purpose of the Study:

  • To investigate whether pancreatic cancer-associated fibroblasts (CAFs) harbor genetic alterations.
  • To determine if CAFs exhibit copy number changes or p53 mutations.

Main Methods:

  • CAFs were cultured from 7 primary pancreatic adenocarcinomas.
  • Genome-wide copy number changes were analyzed using Affymetrix 250K SNP microarrays.
  • p53 protein expression, an indicator of mutation, was assessed via immunohistochemistry in 117 pancreatic ductal adenocarcinomas.

Main Results:

  • Pancreatic CAFs showed no evidence of somatic copy number gains or losses.
  • p53 protein expression was exclusively observed in invasive pancreatic adenocarcinoma cells, not in CAFs.

Conclusions:

  • Pancreatic CAFs do not appear to harbor somatic copy number changes.
  • There is no immunohistochemical evidence of p53 mutations within pancreatic CAFs.
