We developed a sensitive multiplex assay to measure PIWI-interacting RNA (piRNA) expression in male germ cells. This real-time PCR method detects multiple piRNAs from single cells, advancing germ cell development research.

Area of Science:

  • Molecular Biology
  • Genetics
  • Developmental Biology

Background:

  • PIWI-interacting RNAs (piRNAs) are crucial small RNAs in male germ cells.
  • piRNAs interact with PIWI Argonaute proteins like Mili and Miwi.
  • Understanding piRNA function requires sensitive expression analysis methods.

Purpose of the Study:

  • To develop a sensitive, real-time PCR-based multiplex assay for piRNA expression.
  • To enable simultaneous detection of multiple piRNAs from limited RNA samples.
  • To investigate piRNA expression dynamics during early germ cell development.

Main Methods:

  • A novel real-time PCR multiplex assay was designed for piRNA detection.
  • Assay specificity was validated against Northern blot results in adult testis.

Related Experiment Videos

  • The assay's sensitivity was assessed using minimal total RNA input.
  • Main Results:

    • The assay accurately detected piRNA expression in adult testis.
    • It can simultaneously detect at least eight piRNAs using only 10pg of total RNA (single-cell equivalent).
    • The assay demonstrated 5-6 orders of magnitude higher sensitivity than Northern blot.
    • Expression of eight piRNAs was analyzed in mouse primordial germ cells (PGCs) starting at E12.5.

    Conclusions:

    • The developed multiplex piRNA assay is highly sensitive and specific.
    • This method allows for the analysis of piRNA expression from single cells.
    • The assay facilitates the study of piRNA mechanisms and functions in germ cell development.
