Phase Contrast and Differential Interference Contrast Microscopy01:26

Phase Contrast and Differential Interference Contrast Microscopy

Phase-Contrast Microscopes
In-phase-contrast microscopes, interference between light directly passing through a cell and light refracted by cellular components is used to create high-contrast, high-resolution images without staining. It is the oldest and simplest type of microscope that creates an image by altering the wavelengths of light rays passing through the specimen. Altered wavelength paths are created using an annular stop in the condenser. The annular stop produces a hollow cone of...
Interference and Diffraction02:18

Interference and Diffraction

Interference is a characteristic phenomenon exhibited by waves. When two electromagnetic waves interact with their peaks and troughs coinciding, a resulting wave with enhanced amplitude is produced. This is known as constructive interference. In this case, the two waves interacting are in phase with each other.
Super-resolution Fluorescence Microscopy01:37

Super-resolution Fluorescence Microscopy

Super-resolution fluorescence microscopy (SRFM) provides a better resolution than conventional fluorescence microscopy by reducing the point spread function (PSF). PSF is the light intensity distribution from a point that causes it to appear blurred. Due to PSF, each fluorescing point appears bigger than its actual size, and it is the PSF interference of nearby fluorophores that causes the blurred image. Various approaches to achieving higher resolution through SRFM have recently been developed.

    Area of Science:

    • Optics and Photonics
    • Metrology
    • Image Processing

    Background:

    • Projection moiré interferometry is a key technique for non-contact measurement.
    • Accurate generation of reference images is crucial for interferometric precision.
    • Existing methods using cosine-wave and square-wave approaches have limitations in error and processing speed.

    Purpose of the Study:

    • To compare three distinct methods for generating reference images in projection moiré interferometry.
    • To evaluate the performance of an interpolation-based phase-shift technique against conventional methods.
    • To identify the most effective approach for enhancing measurement accuracy and efficiency.

    Main Methods:

    • Comparison of three reference image generation techniques: cosine-wave, square-wave, and interpolation-based phase-shift.
    • Implementation of phase-shifting on actual reference images using the interpolation method.
    • Quantitative analysis of measurement error and processing time for each method.

    Main Results:

    • The interpolation-based phase-shift technique demonstrated a reduction in error exceeding 25%.
    • This novel approach significantly decreased the processing time required for reference image generation.
    • Performance was superior when compared to both cosine-wave and square-wave based methods.

    Conclusions:

    • The interpolation-based phase-shift technique offers a substantial improvement for projection moiré interferometry.
    • This method provides enhanced accuracy and efficiency in generating reference images.
    • It represents a more advanced and practical solution for interferometric measurements.
