The CCR5 Delta32 gene variant increases gallbladder cancer risk, especially in early-onset cases and tobacco users. This genetic susceptibility is independent of gallstones.
Area of Science:
- Genetics
- Oncology
- Immunology
Background:
- Gallbladder inflammation is a known risk factor for gallbladder cancer (GBC).
- Chemokine receptors are involved in immune responses, inflammation, and cancer development.
Purpose of the Study:
- To investigate the association between CCR5 Delta32 polymorphism and genetic susceptibility to GBC.
- To explore how this polymorphism influences GBC risk in relation to factors like gallstones, age of onset, gender, and tobacco use.
Main Methods:
- A case-control study involving 144 GBC patients and 210 healthy controls.
- Genotyping performed using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique.
Main Results:
- A statistically significant difference in CCR5+/Delta32 genotype and CCR5 Delta32 allele distribution was observed between GBC patients and controls.
- The CCR5 Delta32 allele was significantly associated with early-onset GBC (<50 years) and increased risk in tobacco users.
- The association was independent of gallstone presence and showed only marginal gender influence.
Conclusions:
- The CCR5+/Delta32 genotype and CCR5 Delta32 allele represent significant risk factors for GBC.
- This genetic risk is particularly pronounced in individuals with early-onset disease and those who use tobacco.
- CCR5+/Delta32 polymorphism's role in GBC susceptibility is not dependent on gallstone formation.
