Summary
Sharp wave-ripple (SPW-R) oscillations in the hippocampus originate in CA3 and propagate to CA1. Our study shows CA3 activity precisely dictates CA1 network activity during these events.
Area of Science:
- Neuroscience
- Computational Neuroscience
Background:
- Network oscillations facilitate temporal coding by binding neurons into transient assemblies.
- In the hippocampus, place cell sequences represent spatial information and are replayed during sharp wave-ripples (SPW-Rs).
- SPW-Rs originate in CA3 and propagate to CA1, but the precise role of CA3 in defining CA1 activity patterns remains unclear.
Purpose of the Study:
- To investigate the causal relationship between CA3 and CA1 activity during sharp wave-ripple oscillations (SPW-Rs).
- To determine if CA1 network activity is entrained by preceding CA3 neuronal firing patterns.
Main Methods:
- Utilized a mouse hippocampal slice model to study SPW-R propagation between CA3 and CA1.
- Analyzed network and single-cell activity using field potentials and multiunit recordings.
- Employed electrical stimulation in CA3 and antidromic activation in CA1 to probe causal links.
Main Results:
- Demonstrated tight coupling of high-frequency network activity between CA3 and CA1 during SPW-Rs.
- Confirmed that SPW-R propagation preserves precise temporal relationships at network and cellular levels.
- Showed that CA3 stimulation elicits SPW-R-like activity in CA1, whereas CA1 activation does not induce organized oscillations.
Conclusions:
- CA3 activity directly drives and specifies neuronal assembly patterns during SPW-Rs in the hippocampal loop.
- The specificity of neuronal assemblies is maintained with cell-to-cell precision during SPW-R propagation.
- CA3 exerts a dominant role in shaping CA1 network dynamics during memory-related oscillations.
