Sperm Structure and Semen Composition

During ejaculation, males release around 2-5 milliliters of semen, which is a complex mixture of mature sperm and various fluids produced by accessory glands. Summary

Sperm collection methods, post-mortem and electroejaculation, in Iberian red deer show potential for germplasm banking. Both methods yield usable sperm, with electroejaculation offering higher quality and lower abnormality rates for artificial reproductive technologies.

Background:

  • Artificial reproductive technologies (ART) in cervids require foundational data for sperm collection.
  • Existing ART methods for cervids need further development and data collection.

Purpose of the Study:

  • To compare post-mortem (PM) and electroejaculation (EE) sperm collection models in Iberian red deer.
  • To analyze the relationship between stag age and sperm collection parameters.
  • To assess the suitability of collected sperm for germplasm banking and ART.

Main Methods:

  • Collected sperm from 179 wild red deer (PM) and 37 farmed red deer (EE).
  • Recorded testicular/epididymal weight, testicular diameter, sperm quantity, pH, osmolality, motility (CASA), abnormal forms, cytoplasmic droplets, viability, and acrosomal status.
  • Correlated parameters with stag age and compared PM vs. EE methods.

Main Results:

  • Genitalia parameters correlated linearly with stag age.
  • Sperm quantity was similar between PM (2.5x10^9) and EE (3.6x10^9), increasing with age only in PM.
  • EE samples showed significantly different pH and osmolality compared to PM, with higher motility quality, lower abnormal forms, and higher intact acrosomes (97% vs. 89%).

Conclusions:

  • Both post-mortem and electroejaculation sperm collection methods are viable for germplasm banking in Iberian red deer.
  • Electroejaculation yielded higher quality sperm with fewer abnormalities, making it potentially more advantageous for ART.
  • This study provides crucial spermatological data for advancing ART in cervids.
