Total Internal Reflection Fluorescence Microscopy

Total internal reflection fluorescence microscopy or TIRF is an advanced microscopic technique used to visualize fluorophores in samples close to a solid surface with a higher refractive index, such as a glass coverslip. TIRF only allows fluorophores in proximity to the solid surface to be excited. When light from a medium with a lower refractive index (such as air) hits the glass coverslip at a critical angle, the light undergoes total internal reflection stead of passing through the glass.

Summary

Translucent dental posts allow light-cured cement to harden effectively in the cervical and middle root sections. Opaque posts limited cement polymerization, with insufficient hardening observed in the apical regions for both post types.

  • Bovine teeth were prepared and cemented with either translucent (Light-Post) or opaque (Aestheti Post) systems.
  • Root sections were analyzed for cement microhardness using Vickers analysis at cervical, middle, and apical regions.
  • Control groups with only cement were included for comparison.

  • Cement microhardness was insufficient for analysis in the apical regions for both translucent and opaque posts.
  • No statistical differences in microhardness were found between translucent posts (cervical and middle regions) and opaque posts (cervical region).
