Background:

  • Neutrophils play a role in lung injury and adult respiratory distress syndrome (ARDS).
  • Neutrophils can produce toxic substances, including leukotoxin (9,10-epoxy-12-octadecenoate).

Purpose of the Study:

  • To investigate the role of neutrophil-derived leukotoxin in ARDS-associated lung injury.
  • To determine if leukotoxin contributes to coagulation abnormalities in ARDS.

Main Methods:

  • Intravenous injection of leukotoxin in rats to assess lung injury.
  • Detection of leukotoxin in lung lavage fluid from rats exposed to hyperoxia and ARDS patients.
  • Evaluation of coagulation status following leukotoxin administration.

Main Results:

  • Leukotoxin injection caused acute edematous lung injury, characterized by increased lung weight and albumin.
  • Pulmonary capillary endothelial damage and edema were observed via electron microscopy.
  • Leukotoxin was detected in lung lavage of hyperoxia-exposed rats and ARDS patients, correlating with increased neutrophils and albumin.
  • Leukotoxin injection induced disseminated intravascular coagulation (DIC).

  • Neutrophil-produced leukotoxin is a significant factor in ARDS-related lung injury.
  • Leukotoxin contributes to the coagulation abnormalities observed in ARDS patients.
