Summary

This study found that Lactobacillus casei Shirota (LcS) did not prevent peanut allergy in rats. LcS modulated immune responses but did not down-regulate the allergic reaction, challenging its efficacy in allergy prevention.

Background:

  • Probiotics, like Lactobacillus casei Shirota (LcS), are investigated for immune modulation and allergy prevention.
  • The efficacy of probiotics in preventing allergies remains controversial and requires further investigation.

Purpose of the Study:

  • To investigate the effect of Lactobacillus casei Shirota (LcS) on modulating food allergic responses to peanut extract (PE) in Brown Norway (BN) rats.
  • To determine if LcS administration can prevent or down-regulate a peanut-induced allergic response in a rat model.

Main Methods:

  • Brown Norway (BN) rats were sensitized to peanut extract (PE) via oral gavage.
  • Experimental groups received daily oral doses of LcS (1 x 10^9 colony forming units/rat/day).
  • Immune responses, including specific antibody levels (IgG1, IgG2a, IgE), cytokine production (IFN-gamma, IL-4), and basophil counts, were analyzed.

Main Results:

  • LcS had minimal effects on non-sensitized animals.
  • In PE-sensitized rats, LcS shifted the Th1/Th2 ratio towards Th2 dominance and increased PE-specific IgG2a and basophil counts.
  • While LcS increased Th1 and Th2 cytokines (IFN-gamma and IL-4), IL-4 levels were predominantly elevated in spleen and mesenteric lymph node cells.

Conclusions:

  • The study does not support the hypothesis that LcS down-regulates food allergic responses in this rat model.
  • LcS modulated immune responses but did not prevent or reduce the severity of peanut allergy in BN rats.
  • Further research is needed to understand the complex interactions between probiotics and the immune system in allergy development.
