Summary

Pancreatic progenitor cells exist in a hierarchy, from rare multipotent progenitors to abundant monopotent ones. This model explains how the pancreas regenerates after injury or during homeostasis.

Area of Science:

  • Cell Biology
  • Developmental Biology
  • Gastroenterology

Background:

  • Past pancreatic progenitor cell research relied heavily on histological analyses.
  • Recent studies suggest the existence of diverse pancreatic progenitor cells with varying lineage potentials in developing and adult pancreases.

Purpose of the Study:

  • To propose a hierarchical model for pancreatic progenitor cells.
  • To explain progenitor cell activation based on injury severity and physiological state.
  • To differentiate functional roles of ductal and endocrine progenitor cells.

Main Methods:

  • Review of recent genetic labeling, cell tracing, single-cell analyses, and colony assays.
  • Integration of findings on progenitor cell frequency and lineage potential.

  • Functional analysis of endocrine progenitor cell activity.
  • Main Results:

    • Pancreatic progenitor cells are proposed to be organized in a hierarchy, with rare, primitive multipotent progenitors at the top and abundant monopotent progenitors at the bottom.
    • This hierarchy explains differential progenitor cell activation in response to varying injury levels and physiological conditions.
    • Ductal cells are functionally heterogeneous, and endocrine progenitor activity is best assessed functionally, not histologically.

    Conclusions:

    • A hierarchical model of pancreatic progenitor cells provides a framework for understanding pancreatic regeneration and homeostasis.
    • Functional assessment is crucial for determining endocrine progenitor cell activity.
    • Future research should focus on the functional heterogeneity of ductal cells and the precise roles of different progenitor populations.
