Summary

A new method accurately measures oxygen diffusion in cell-laden gel beads. Oxygen diffusivity is unaffected by cell density or type, with Ca-alginate beads showing superior performance.

Background:

Accurate measurement of oxygen diffusion is critical for understanding cellular processes in immobilized systems.
Existing methods for determining oxygen diffusion coefficients in gel beads can be complex and time-consuming.
Viable cells within gel matrices can significantly alter oxygen transport dynamics.

Purpose of the Study:

To develop and validate a simple, steady-state method for measuring the effective diffusion coefficient of oxygen (D(e)) in gel beads containing viable cells.
To investigate the influence of cell density, cell type, and cell viability on oxygen diffusivity within alginate gel beads.
To compare oxygen diffusion in calcium (Ca) and barium (Ba)-alginate gel beads and relate it to cell distribution.

A steady-state method was employed to measure the effective diffusion coefficient of oxygen.
Experiments were conducted using Ca- and Ba-alginate gel beads entrapping Saccharomyces cerevisiae and Pseudomonas ovalis.
Oxygen concentration profiles within single gel beads were analyzed and compared with cell distribution.

Main Results:

The developed method achieved an accuracy of +/-7% for measuring D(e).
Oxygen diffusivity was found to be independent of cell density (0-30 g/L), cell type, and cell viability.
Ca-alginate gel beads exhibited superior oxygen diffusivity compared to Ba-alginate beads, approaching that of the surrounding liquid.
Oxygen concentration profiles aided in estimating the thickness of the cell layer within gel beads.

Conclusions:

The proposed steady-state method provides a reliable and simple approach for quantifying oxygen diffusion in cell-laden gel beads.
Cellular factors do not significantly impact oxygen diffusion rates in these gel systems.
Ca-alginate beads are more efficient for oxygen transport, and microgel bead dimensions can optimize effectiveness factors for enhanced cellular performance.
