Thyroid hormone receptors (TR) isoforms are expressed in human astrocytomas. Their expression levels correlate with tumor grade, suggesting potential for hormonal therapy in brain tumors.
Area of Science:
- Neuroscience
- Endocrinology
- Oncology
Background:
- Thyroid hormone is crucial for brain development, influencing astrocyte maturation.
- Aberrant expression of thyroid hormone receptor (TR) isoforms is implicated in tumorigenesis.
- Limited research exists on TR isoform expression in human astrocytomas.
Purpose of the Study:
- To investigate the expression of TR isoforms in human astrocytomas.
- To correlate TR isoform expression with astrocytoma malignancy grade.
Main Methods:
- Reverse transcription-polymerase chain reaction (RT-PCR) was employed.
- Expression of human TR isoforms was analyzed in 34 astrocytoma samples.
Main Results:
- TR isoform expression was detected in human astrocytomas.
- TRalpha1 and TRalpha2 expression frequency decreased with increasing malignancy grade (P=.005, P=.043).
- TRbeta1 expression frequency significantly increased with astrocytoma malignancy grade (P=.017).
Conclusions:
- This study is the first to demonstrate TR isoform expression in human astrocytomas.
- TR isoform expression levels correlate with astrocytoma malignancy grading.
- Findings suggest potential for hormonal therapy in brain tumors based on TR expression.
