Summary

Microcrystalline cellulose (MCC) starters yield the best results for active pellet formation due to their insolubility and mechanical strength. Sucrose starters dissolved, leading to lower yields and agglomerates in drug layering processes.

Main Results:

  • Microcrystalline cellulose (MCC) starters resulted in the highest yield (85.66-89.41%) due to their water insolubility and mechanical integrity.
  • Sucrose starters showed lower yields (76.2%) and formed undesirable agglomerates because of partial dissolution in the drug solvent.
  • All tested pellet formulations exhibited favorable flow properties and drug content ranging from 82.4% to 94.5% of the theoretical amount.

Conclusions:

  • Inert starter core composition critically affects active pellet yield and quality.
  • MCC is a superior starter material for drug layering via Wurster coating compared to sucrose, offering better processability and product consistency.
