Summary

Stem cells from human exfoliated deciduous teeth (SHED) can form dental pulp-like tissue in vivo. These multipotent stem cells differentiate into odontoblast-like and endothelial-like cells, showing promise for pulp regeneration.

Background:

Stem cells from human exfoliated deciduous teeth (SHED) are multipotent.
The potential of SHED to regenerate dental pulp tissue in vivo remains unexplored.

Purpose of the Study:

To evaluate the in vivo formation of dental pulp-like tissue using SHED seeded in biodegradable scaffolds.
To assess the differentiation capacity of SHED within a tooth slice scaffold in immunodeficient mice.

Main Methods:

SHED were seeded onto biodegradable scaffolds within human tooth slices.
Scaffolds were transplanted into immunodeficient mice.
Tissue morphology, cellularity, ultrastructure, and cell differentiation were analyzed.

Main Results:

Engineered tissue exhibited architecture and cellularity similar to physiologic dental pulp.
SHED differentiated into odontoblast-like cells, confirmed by dentin sialoprotein expression.
SHED also differentiated into endothelial-like cells, forming blood vessel linings.

Conclusions:

SHED can successfully generate dental pulp-like tissue in vivo.
Exfoliated deciduous teeth are a promising source of stem cells for dental pulp tissue engineering.
SHED demonstrate multipotency, differentiating into both odontogenic and endothelial lineages.
