Area of Science:

  • Developmental Biology
  • Cell Biology
  • Microscopy Techniques

Background:

  • Drosophila pupal wing development involves complex epithelial and glial morphogenesis.
  • Understanding these dynamic events is crucial for cell biologists.
  • Existing protocols may limit detailed visualization or live imaging.

Purpose of the Study:

  • To establish optimized protocols for preparing Drosophila pupal and prepupal wings for immunofluorescence microscopy.
  • To enable detailed visualization of wing epithelial architecture, including planar cell polarity.
  • To provide a method for whole-mount live imaging of pupal wings, focusing on glial cell behavior.

Main Methods:

  • Protocol development for immunofluorescence microscopy of Drosophila pupal (P) and prepupal (PP) wings.

  • Optimization of sample preparation for visualizing epithelial structures.
  • Development of a whole-mount live imaging protocol for P wings.
  • Main Results:

    • Successfully visualized polarized cortical domains of planar cell polarity proteins in wing epithelial architecture.
    • Enabled live imaging of glial cell migration and proliferation along wing sensory nerves.
    • Demonstrated the utility of the developed protocols for studying dynamic developmental processes.

    Conclusions:

    • The presented protocols significantly advance the study of Drosophila wing development.
    • These methods facilitate detailed analysis of epithelial organization and cell dynamics.
    • The protocols are valuable tools for cell biologists investigating morphogenesis and cell migration.
