Summary

Small cell lung cancer may originate from multipotent airway stem cells. Researchers found shared stem cell marker expression (podocalyxin-like protein 1 and Bmi-1) between small cell lung cancers and developing proximal bronchial cells.

  • Analysis of podocalyxin-like protein 1 promoter methylation status in SCLC.
  • Main Results:

    • 87.5% of SCLC samples expressed podocalyxin-like protein 1 (PODXL-1), a stem cell marker.
    • 98.4% of SCLC samples ubiquitously expressed Bmi-1, a self-renewal factor.
    • PODXL-1 was detected in clustered epithelial cells of developing proximal bronchi and trachea, but not in normal adult bronchial epithelium.

    Conclusions:

    • Shared expression of stem cell markers (PODXL-1, Bmi-1) between SCLC and developing proximal bronchial epithelial cells suggests a common histogenetic origin.
    • Epigenetic regulation, indicated by demethylated CpG islands in the PODXL-1 promoter, may play a role in SCLC development.
    • These findings support the hypothesis that SCLC may arise from a multipotent proximal airway epithelial precursor cell.
