Summary

Seminal plasma hinders boar sperm capacitation after cryopreservation. However, adding seminal plasma back to frozen-thawed semen did not impact fertility outcomes in sows.

Purpose of the Study:

  • To investigate the effects of seminal plasma on the functional status of cryopreserved boar sperm.
  • To determine if seminal plasma influences sperm capacitation and fertility after thawing.

Main Methods:

  • Boar sperm were evaluated for functional changes after processing and cryopreservation.
  • The impact of seminal plasma addition to frozen-thawed semen was assessed.
  • Sperm presence in the utero-tubal reservoir and farrowing rates were measured.

Main Results:

  • Seminal plasma was found to prevent or reverse capacitation in frozen-thawed boar sperm.
  • Supplementation with 10% seminal plasma did not significantly alter sperm numbers in the utero-tubal sperm reservoir.
  • Farrowing rates were not significantly affected by the addition of seminal plasma to cryopreserved semen.

Conclusions:

  • Seminal plasma has a detrimental effect on boar sperm capacitation post-cryopreservation.
  • Despite affecting sperm function in vitro, seminal plasma supplementation does not negatively impact fertility parameters in vivo.
  • These findings suggest a complex interaction between seminal plasma, sperm physiology, and fertility in pigs.
