Purpose of the Study:

  • To investigate if percutaneous and surgical liver biopsies, both histologically normal, exhibit different gene expression patterns.
  • To determine the impact of tissue acquisition method on control gene expression profiles.
  • To identify specific genes whose expression differs between percutaneous and surgical liver biopsies.

Main Methods:

  • Comparison of messenger RNA (mRNA) expression of 240 selected genes.
  • Real-time quantitative reverse-transcription polymerase chain reaction (RT-qPCR).
  • Analysis of 14 percutaneous liver biopsies (Group A) versus 14 surgical liver biopsies (Group B) from histologically normal livers.

Main Results:

  • Significant differences in the expression of 26 out of 240 genes were observed between the two groups.
  • 23 genes were upregulated in the percutaneous group (Group A), and 3 genes were downregulated.
  • Eight genes, including PAI1, THBS1, IL8, PTGS2, CXCR4, JUN, and FOS, showed perfect discrimination between the groups. IL8 expression varied in chronic hepatitis C samples based on control type.

Conclusions:

  • Histologically normal liver tissue obtained via percutaneous or surgical methods displays distinct gene expression profiles.
  • The choice of control tissue in gene expression studies is critical.
  • Inadequate selection of normal controls can lead to discordant research findings.
