The semi-quantitative 2-deoxyglucose method for measuring local cerebral glucose utilization (LCMRglu) is less valid than the quantitative approach. A new analysis improves LCMRglu estimation and broadens experimental applications.
Area of Science:
- Neuroscience
- Biochemistry
- Medical Imaging
Background:
- The quantitative [(14)C]-2-deoxyglucose autoradiography technique measures local cerebral glucose utilization (LCMRglu).
- This quantitative method requires frequent arterial blood sampling, limiting its application.
- A semi-quantitative approach bypasses serial sampling but has questionable validity.
Purpose of the Study:
- To evaluate the validity of the semi-quantitative LCMRglu method compared to the quantitative approach.
- To address limitations in determining metabolic stability within the semi-quantitative method.
- To develop a novel analytical technique to improve LCMRglu estimation using the semi-quantitative method.
Main Methods:
- Comparison of quantitative and semi-quantitative [(14)C]-2-deoxyglucose autoradiography for LCMRglu measurement.
- Analysis of plasma [(14)C] and glucose concentrations from terminal blood samples.
- Development and application of a novel analytical method for index of LCMRglu (iLCMRglu) estimation.
Main Results:
- The semi-quantitative method shows reduced validity in LCMRglu measurement compared to the quantitative method.
- The ability to determine metabolic stability in the standard region is compromised in the semi-quantitative approach.
- The novel analytical technique enhances the validity of LCMRglu estimation via the semi-quantitative method.
Conclusions:
- The developed analytical technique improves the accuracy of the semi-quantitative LCMRglu method.
- This novel approach broadens the experimental applicability of the semi-quantitative 2-deoxyglucose autoradiography.
- Adherence to methodological constraints like normoglycaemia is crucial for valid iLCMRglu estimation.
