Leaky Scanning

During most eukaryotic translation processes, the small 40S ribosome subunit scans an mRNA from its 5' end until it encounters the first start AUG codon. The large 60S ribosomal subunit then joins the smaller one to initiate protein synthesis. Summary

Canine procalcitonin messenger RNA (mRNA) was measured using qRT-PCR in ill and healthy dogs. Procalcitonin mRNA was higher in ill dogs, suggesting it is an acute phase protein, with potential prognostic value.

Background:

  • Procalcitonin is an acute phase protein used in human medicine for infection and prognosis.
  • Canine procalcitonin has been sequenced, but its diagnostic or prognostic utility in dogs is unassessed.

Purpose of the Study:

  • To develop and validate a quantitative reverse transcription polymerase chain reaction (qRT-PCR) assay for canine procalcitonin mRNA.
  • To assess procalcitonin mRNA expression as a diagnostic and prognostic marker in ill dogs.

Main Methods:

  • A qRT-PCR assay was developed for canine procalcitonin mRNA.
  • Whole blood samples from 70 dogs (11 healthy, 59 ill) were analyzed.
  • Dogs were categorized into inflammatory, infectious, neoplastic, other diseases, and healthy control groups.

Main Results:

  • Procalcitonin mRNA expression was significantly higher (P < 0.001) in all ill dogs compared to healthy controls.
  • No significant differences in procalcitonin mRNA expression were found between different ill dog groups.
  • No correlations were observed between procalcitonin mRNA levels and hematological measures.

Conclusions:

  • Canine procalcitonin functions as an acute phase protein.
  • Procalcitonin mRNA expression shows potential prognostic value, as changes aligned with clinical improvement or deterioration in a subset of dogs.
