Summary

Friend leukemia integration site 1 (Fli-1) is a nuclear marker found in melanoma cells. Higher Fli-1 expression in malignant melanoma correlates with more aggressive tumor characteristics.

Area of Science:

  • Oncology
  • Molecular Biology
  • Immunohistochemistry

Background:

  • Friend leukemia integration site 1 (Fli-1) is a nuclear marker for endothelial differentiation.
  • Fli-1 is expressed in leukocytes and has been identified in melanoma cells.

Purpose of the Study:

  • To investigate the expression of Fli-1 in melanoma.
  • To determine the association between Fli-1 expression and melanoma aggressiveness.

Main Methods:

  • Immunohistochemistry was used to evaluate Fli-1 expression in 97 melanoma tissue sections (69 primary, 28 metastatic).
  • Western blot and immunocytochemistry were performed on five melanoma cell lines.
  • Spearman correlation was used to analyze the relationship between Fli-1 expression and clinicopathological features.

Main Results:

  • Fli-1 expression was detected in all evaluated melanoma cell lines.
  • Fli-1 expression was significantly higher in metastatic melanomas compared to primary tumors (p=0.041).
  • Fli-1 expression positively correlated with Ki-67 expression (p=0.022) and the presence of ulceration in primary tumors (p=0.030).

Conclusions:

  • Fli-1 expression in malignant melanoma is associated with biologically aggressive tumors.
  • Fli-1 may serve as a potential biomarker for melanoma progression and aggressiveness.
