Hepatitis C virus (HCV) treatment with interferon-alpha(2) (IFN-alpha(2)) induces type I IFN regulated genes in blood cells, but not in the liver. This suggests HCV clearance mechanisms may not involve hepatic type I IFN gene activation.

- Expression of MxA and PKR (type I IFN-regulated genes) was quantified in PBMCs and liver samples.

- IFN-alpha(2) treatment induced type I IFN-regulated genes (MxA, PKR) in PBMCs.
- No significant induction of these type I IFN-regulated genes was observed in the liver.
- Only one identified upregulated gene during effective treatment was potentially type I IFN-responsive.
Conclusions:
- IFN-alpha(2) treatment induces type I IFN-regulated genes in PBMCs of HCV patients.
- This induction does not appear to occur in the liver.
- HCV viral clearance during IFN-alpha(2) therapy may be independent of hepatic type I IFN-regulated gene activation.
