Multiple myeloma (MM) causes bone destruction by increasing osteoclast activity and decreasing bone formation. New treatments targeting these processes show promise for managing MM bone disease.
Area of Science:
- Oncology
- Bone Biology
- Pharmacology
Background:
- Multiple myeloma (MM) frequently causes osteolytic bone lesions due to increased osteoclast activity and suppressed osteoblast function.
- This imbalance leads to significant bone destruction and impaired bone formation in affected patients.
Purpose of the Study:
- To review the pathophysiology of bone remodeling in multiple myeloma.
- To discuss emerging therapeutic strategies for treating MM-related bone disease.
Main Methods:
- Literature review of the pathophysiology of MM bone disease.
- Discussion of novel therapeutic targets and agents currently in or nearing clinical trials.
Main Results:
- Advances in understanding MM pathogenesis have identified novel therapeutic targets.
- Several new agents targeting MM bone disease are progressing towards clinical trials.
Conclusions:
- Combination therapies including immunomodulatory drugs (IMiDs), bortezomib, and novel anti-resorptive agents are expected to be effective.
- These combinations are anticipated in clinical trials soon, though their ability to fully 'heal' bone lesions remains uncertain.
