UV–Vis Spectrometers01:14

UV–Vis Spectrometers

The absorbance of UV and visible (UV–visible) radiations is measured using a UV–visible spectrophotometer. Deuterium lamps, which emit UV radiation, and tungsten lamps, which produce radiation in the visible region, are used as light sources in UV–visible spectrophotometers. A monochromator or prism is used for diffraction grating, i.e., to split the incoming radiation into different wavelengths. A system of slits is used to focus the desired wavelength on the sample cell. Samples for...

Ultraviolet and Visible (UV–Vis) Spectroscopy: Overview

Ultraviolet–visible (UV–visible or UV–Vis) spectroscopy is an analytical technique that investigates the interaction between matter and UV–Vis light within the electromagnetic spectrum. This method is widely used for its versatility, simplicity, and relatively quick data acquisition, making it valuable for both qualitative and quantitative analysis. When UV–Vis radiation passes through a material,  molecules absorb light depending on the energy required for electronic transitions. As a result...

Atomic Absorption Spectroscopy: Lab

For AAS measurements, samples must be introduced as clear solutions, often requiring extensive preliminary treatment to dissolve materials like soils, animal tissues, and minerals. Common methods for sample preparation include treatment with hot mineral acids, wet ashing, combustion in closed containers, high-temperature ashing, or fusion with reagents.
Solutions containing organic solvents, such as low-molecular-mass alcohols, esters, or ketones, enhance absorbances by increasing nebulizer...
Microbial Bioremediation of Uranium01:25

Microbial Bioremediation of Uranium

Microorganisms play a critical role in the transformation and immobilization of uranium in contaminated environments through four main pathways: bioreduction, biosorption, bioaccumulation, and biomineralization. These mechanisms reduce uranium’s toxicity and prevent its migration through groundwater systems, offering sustainable approaches for in situ bioremediation.Bioreduction of UraniumBioreduction is driven by anaerobic bacteria such as certain strains of Geobacter and Shewanella, which use...

Area of Science:

  • Analytical Chemistry
  • Inorganic Chemistry

Background:

  • Accurate determination of uranium is crucial for environmental monitoring and nuclear industry applications.
  • Existing spectrophotometric methods for uranium often lack sufficient sensitivity or require complex procedures.

Purpose of the Study:

  • To develop a highly sensitive spectrophotometric method for uranium (U) determination.
  • To investigate the complexation reaction between uranium, Pyrogallol Red (PGR), and cetyltrimethylammonium bromide (CTAB).

Main Methods:

  • Spectrophotometric analysis of uranium using Pyrogallol Red (PGR) as a chromogenic agent.
  • Inclusion of cetyltrimethylammonium bromide (CTAB) to enhance the sensitivity of the color reaction.
  • Characterization of the ternary complex formed, including its composition and optimal reaction conditions (pH 5.6).

Main Results:

  • A sensitive blue ternary complex of uranium with PGR and CTAB was formed, with maximum absorbance at 580 nm.
  • The method demonstrated adherence to Beer's law over a uranium concentration range of 0.24–19.0 ppm.
  • High molar absorptivity (3.3 x 10^4 L.mol^-1.cm^-1) was achieved at 620 nm, indicating excellent sensitivity.

Conclusions:

  • The developed spectrophotometric method offers a sensitive and reliable approach for uranium quantification.
  • The presence of CTAB significantly enhances the sensitivity of the uranium-PGR reaction.
  • The study also proposes a method for evaluating the stability constants of such ternary complexes.
