Summary

A new UR cell line effectively produces replication-competent adenovirus (RCA)-free adenovirus vectors, unlike the traditional 293 cell line. This advancement enhances the safety of gene therapy and vaccination delivery systems.

Area of Science:

  • Molecular Biology
  • Virology
  • Biotechnology

Background:

  • Recombinant adenoviruses (Ad) are vital for gene therapy and vaccination delivery.
  • The human embryonic kidney 293 cell line, commonly used for Ad production, poses a risk of generating replication-competent adenoviruses (RCA).

Purpose of the Study:

  • To develop a novel cell line for producing RCA-free adenovirus vectors.
  • To ensure the safety and stability of adenovirus vectors for therapeutic applications.

Main Methods:

  • Construction of a shuttle plasmid encoding the essential E1A/E1B sequence for adenovirus replication.
  • Transfection of the shuttle plasmid into human embryonic lung HEL 299 cells to create the UR cell line.
  • Confirmation of E1A/E1B sequence integration and protein expression in the UR cells.

  • Assessment of RCA generation in viruses produced by UR cells compared to 293 cells over multiple passages.
  • Main Results:

    • The UR cell line was successfully established and confirmed to contain the E1A/E1B sequence with active protein expression.
    • Adenovirus vectors produced using the UR cell line remained free of RCA after ten passages.
    • Adenovirus produced in the traditional 293 cell line generated RCA by the fourth passage.

    Conclusions:

    • The UR cell line provides a stable and effective platform for producing RCA-free adenovirus vectors.
    • UR cells yield comparable virus titers to 293 cells without compromising safety.
    • This new cell line significantly mitigates the risk of RCA formation, enhancing the safety profile of adenovirus-based gene therapies and vaccines.
