Clinical Applications of Epidermal Stem Cells01:19

Clinical Applications of Epidermal Stem Cells

Epidermal stem cells (EpiSCs) are mainly located at the basal layer of the epidermis. These cells repair minor injuries of the skin and replace dead skin cells. However, EpiSCs’ cannot heal severe wounds such as major burns or those from diabetes or hereditary disorders. In such cases, culturing the epidermal stem cells from the patient is possible and has yielded successful treatment options, such as laboratory-grown skin grafts. These grafts are synthesized using a patient’s own EpiSCs...

Summary

Cultivated epithelial transplantation offers a novel surgical approach for severe ocular surface disorders. Autologous cultivated oral mucosal epithelial transplantation (COMET) provides a viable solution for bilateral cases, reconstructing the ocular surface with non-ocular cells.

Area of Science:

  • Ophthalmology
  • Regenerative Medicine
  • Tissue Engineering

Background:

  • Severe ocular surface disorders pose significant challenges to vision.
  • Current treatments like cultivated corneal epithelial transplantation are effective but limited to unilateral cases.
  • A need exists for strategies addressing bilateral ocular surface conditions.

Purpose of the Study:

  • To introduce autologous cultivated oral mucosal epithelial transplantation (COMET) as a treatment for severe, bilateral ocular surface disorders.
  • To highlight COMET's potential for ocular surface reconstruction using non-ocular cells.

Main Methods:

  • Utilizing tissue-engineered epithelial sheets for rapid corneal surface coverage.
  • Employing autologous oral mucosal cells for transplantation.

Main Results:

  • Cultivated epithelial sheets reduce inflammation and postoperative complications.
  • COMET enables reconstruction of the ocular surface in bilateral cases.

Conclusions:

  • Cultivated epithelial transplantation is a promising modality for ocular surface disorders.
  • COMET expands treatment options for severe, bilateral ocular surface diseases, offering a new pathway for reconstruction.
