Summary

B cell activation requires B cell receptor (BCR) engagement and T-cell help, delivered either directly or indirectly. This study demonstrates in vitro B cell helper assays to understand T-cell dependent B cell responses.

Background:

  • B lymphocyte activation, proliferation, and differentiation into memory B cells and plasma cells depend on B cell receptor (BCR) engagement and T-cell help.
  • T-cell help can be provided in a cognate manner (recognizing MHC-peptide complexes on B cells) or a non-cognate/bystander manner (activated by alternative mechanisms).

Purpose of the Study:

  • To demonstrate and validate in vitro B cell helper assays for studying T-cell dependent B cell activation.
  • To provide reproducible experimental models for deciphering the molecular mechanisms underlying B cell responses.
