Immunocytochemistry and Immunohistochemistry01:22

Immunocytochemistry and Immunohistochemistry

Immunocytochemistry (ICC) and immunohistochemistry (IHC) are techniques that use antibodies to check for specific proteins or antigens in a sample. The technique was first published by Albert Coons in 1941 to detect the presence of pneumococcal antigen in tissue sections from mice infected with Pneumococcus. Immunocytochemistry helps localization of proteins or antigens in individual cells like blood cells, stem cells, etc., while immunohistochemistry does the same for tissue samples.

Area of Science:

  • Surgical Pathology
  • Immunohistochemistry
  • Oncology

Background:

  • D2-40 is a monoclonal antibody targeting human podoplanin.
  • Podoplanin is a mucoprotein expressed in lymphatic endothelial cells.
  • D2-40 immunoexpression is observed in diverse neoplasms.

Purpose of the Study:

  • To review the expanding applications of D2-40 immunohistochemistry.
  • To update on D2-40's utility in surgical pathology.
  • To highlight D2-40's role in diagnosing various tumors.

Main Methods:

  • Literature review of studies reporting D2-40 immunoexpression.
  • Analysis of D2-40's diagnostic utility in different tumor types.
  • Synthesis of current findings on D2-40 applications.

Main Results:

  • D2-40 immunoexpression is documented in lymphovascular neoplasms (e.g., lymphangioma, Kaposi sarcoma).
  • D2-40 is also found in nonvascular neoplasms (e.g., epithelioid mesothelioma, seminoma).
  • Recent reports show D2-40 expression in adrenal cortical tumors, schwannomas, and skin adnexal tumors.

Conclusions:

  • D2-40 immunohistochemistry is a valuable tool in surgical pathology.
  • The diagnostic applications of D2-40 are broad and increasing.
  • D2-40 aids in the identification of a wide spectrum of neoplastic conditions.
