The TGF-β signaling pathway regulates cell growth, differentiation, adhesion, motility, and development. TGF-β ligands that induce TGF-β signaling are synthesized in their latent form. Transforming growth factor (TGF) beta1 expression is higher in high-risk ameloblastomas. This protein may promote tumor invasion in recurrent ameloblastoma cases.

Background:

  • Ameloblastomas (AMs) are odontogenic tumors with varying recurrence risks.
  • Transforming growth factor (TGF) beta1 plays complex roles in tumor progression.

Purpose of the Study:

  • To compare TGF-beta1 expression in ameloblastomas with different recurrence risks.
  • To investigate the association between TGF-beta1 and ameloblastoma invasiveness.

Main Methods:

  • Immunohistochemistry was used to evaluate TGF-beta1 expression.
  • 29 ameloblastoma cases were classified into low-risk (unicystic, peripheral) and high-risk (solid) groups.

Main Results:

  • Higher TGF-beta1 positivity was observed in the stromal cells of high-risk ameloblastomas compared to low-risk ones (P = 0.0308).
  • No significant differences in TGF-beta1 immunoreactivity were found in vessels or odontogenic epithelium between groups.

Conclusions:

  • Elevated TGF-beta1 expression in stromal cells correlates with a higher risk of recurrence in ameloblastomas.
  • TGF-beta1 may contribute to the invasive phenotype of ameloblastomas in later stages.
