Abnormal Proliferation02:23

Abnormal Proliferation

Under normal conditions, most adult cells remain in a non-proliferative state unless stimulated by internal or external factors to replace lost cells. Abnormal cell proliferation is a condition in which the cell's growth exceeds and is uncoordinated with normal cells. In such situations, cell division persists in the same excessive manner even after cessation of the stimuli, leading to persistent tumors. The tumor arises from the damaged cells that replicate to pass the damage to the daughter...
Negative Regulator Molecules01:23

Negative Regulator Molecules

Positive regulators allow a cell to advance through cell cycle checkpoints. Negative regulators have an equally important role as they terminate a cell’s progression through the cell cycle—or pause it—until the cell meets specific criteria.
The Intrinsic Apoptotic Pathway01:31

The Intrinsic Apoptotic Pathway

Internal cellular stress, such as cellular injury or hypoxia, triggers intrinsic apoptosis. The B-cell lymphoma 2 (Bcl-2) family of proteins are the primary regulators of the intrinsic apoptotic pathway. For example, during DNA damage, checkpoint proteins, such as Ataxia Telangiectasia Mutated (ATM protein) and Checkpoints Factor-2 (Chk2) proteins, are activated. These proteins phosphorylate p53 which further activates pro-apoptotic proteins, such as Bax, Bak, PUMA, and Noxa, and inhibits...

Summary

p63 protein expression in diffuse large B-cell lymphoma (DLBCL) is not linked to specific subtypes but is associated with improved disease-free survival (DFS) in high-risk patients, suggesting a potential prognostic role.

Area of Science:

  • Oncology
  • Molecular Biology
  • Immunology

Background:

  • p63 is a p53 homologue with various protein isoforms, some of which can influence apoptosis and gene transactivation.
  • p63 expression in germinal center lymphocytes suggests a potential role in lymphoma development.
  • The prognostic significance of p63 in diffuse large B-cell lymphoma (DLBCL) survival is not well-established.

Purpose of the Study:

  • To investigate the correlation between p63 protein expression and CD10, Bcl-6, and IRF4 antigen expression in DLBCL.
  • To determine if p63 serves as a predictive marker for overall survival in high-risk DLBCL patient cohorts.

Main Methods:

  • Retrospective immunohistochemical (IHC) analysis of CD10, Bcl-6, IRF4, and p63 expression in 73 high-intermediate and high-risk DLBCL samples.
  • Classification of DLBCL samples into Germinal Center B-cell-like (GCB) and Activated B-cell-like (ABC) subtypes based on antigen expression.
  • Comparison of p63 expression levels with DLBCL subtypes and patient survival rates.

Main Results:

  • p63 expression was observed in malignant cells of 11 patients.
  • No significant correlation was found between p63 expression and GCB-like or ABC-like DLBCL subtypes.
  • Patients with p63-positive DLBCL demonstrated significantly better disease-free survival (DFS) compared to p63-negative patients (p = 0.01).

Conclusions:

  • p63 positivity in high-intermediate and high-risk DLBCL is associated with improved disease-free survival.
  • p63 may represent a favorable prognostic marker in specific DLBCL patient groups.
