    Summary

    Resting hematopoietic stem cells can be activated by cyclic guanosine monophosphate (cGMP). This finding suggests cGMP plays a key role in triggering stem cell proliferation for blood formation.

    Background:

    • The activation mechanism of resting hematopoietic stem cells (HSCs) remains unclear.
    • Cyclic nucleotide levels are hypothesized to influence HSC activation.

    Purpose of the Study:

    • To investigate the role of cyclic nucleotides in HSC activation.
    • To determine if cyclic guanosine monophosphate (cGMP) can stimulate the formation of granulocyte colonies in vitro.

    Main Methods:

    • Examined the effect of various nucleotides and synthesis stimulators on in vitro granulocyte colony formation.
    • Assessed the sensitivity of activated cells to thymidine suicide and colony-stimulating activity.

    Main Results:

    • Physiologic concentrations of cGMP enhanced granulocyte-macrophage colony formation with colony-stimulating activity.
    • cGMP-activated cells were resistant to thymidine suicide.
    • Colony-stimulating activity alone did not activate these cells.

    Conclusions:

    • cGMP can trigger normally resting HSCs into a proliferative state.
    • cGMP-mediated activation is distinct from activation by colony-stimulating activity alone.
    • These findings elucidate a novel pathway for HSC activation.
